Published evidence reports the existence of two routes for the transformation of 7-dehydrocholecalciferol into previtamin D3: a photochemical route with the participation of UVB photons and another route that occurs in the darkness. Several reports appears to support the presence of these two routes in some mammals, birds, nonvascular plants (e.g.: mosses), vascular plants (e.g.: angiosperms) and lichens. The reviewed evidence suggests that in the darkness, the synthesis of vitamin D3 follows the same scheme of the photochemical pathway, but at a reduced rate respect to the synthesis under UVB radiation. The process of vitamin D synthesis in the dark, then, may be taken as an insurance for survival, at least for mammals and birds. The low rate of the synthesis of vitamin D3 in the absence of light produce low concentrations of vitamin D3 metabolites in plasma. Long term survival under these circumstances might be possible through upregulation of vitamin D receptors (VDRs). In mole rats (South African rodents that live in the dark in underground tunnels), the reduced rate of vitamin D3 synthesis produce low levels of plasma vitamin D3 and their metabolites 25(OH) D3 and 1α,25OH2 D3 . The fact that Kd and ßmax of the complex 1α,25(OH)2 D3 -VDR from the intestinal mucosa, kidneys and the Harderian glands of the mole rat Heterocephalus glaber are significantly different in each one of these tissues, is interpreted as an indicator that the VDRs are, in each tissue, adapted to the maintenance of normal physiological functions. Varios trabajos publicados han informado que existen dos mecanismos para la transformación de 7-dehidrocolecalciferol en previtamina D3 : uno iniciado con el auxilio de fotones UVB y un segundo que ocurre en la oscuridad, sin el auxilio de radiación ultravioleta. Una serie de publicaciones contienen información que apoya la presencia de estos dos mecanismos en mamíferos, pájaros, plantas no vasculares (musgos), vasculares (angiospermas) y líquenes. La evidencia revisada sugiere que, en la oscuridad, la síntesis de vitamina D3 sigue el mismo esquema que la ruta fotoquímica y generalmente ocurre a una tasa reducida respecto de la síntesis bajo irradiación ultravioleta. La operación de la síntesis de vitamina D3 en la oscuridad, por lo menos para mamíferos y pájaros, puede tomarse como un reaseguro de sobrevida. La reducida tasa de síntesis de vitamina D en ausencia de luz produce bajas concentraciones en plasma de los metabolitos de la vitamina. La sobrevida saludable en estas condiciones sería posible mediante upregulation de los receptores. En las ratas topo Heterocephalus glaber (roedores sudafricanos que viven en permanente oscuridad, en túneles subterráneos), la reducida tasa de síntesis de vitamina D3 es la causa de los bajos niveles plasmáticos de la vitamina y sus metabolitos: 25(OH)D3 y 1α,25OH2 D3 . El hecho de que el Kd y ßmax del complejo 1α,25(OH)2 D3 -Vitamina-D-Receptor del intestino, riñón y glándulas de Harder de Heterocephalus glaber sean significativamente diferentes entre sí indicaría que los receptores se han modificado para mantener funciones fisiológicas normales en cada tejido. BACKGROUND: Epanorin (EP) is a secondary metabolite of the Acarospora lichenic species. EP has been found in lichenic extracts with antimicrobial activity, and UV-absorption properties have been described for closely related molecules; however, its antiproliferative activity in cancer cells has not yet been explored. It has been hypothesized that EP inhibits cancer cell growth. MCF-7 breast cancer cells, normal fibroblasts, and the non-transformed HEK-293 cell line were exposed to increasing concentrations of EP, and proliferation was assessed by the sulforhodamine-B assay. RESULTS: MCF-7 cells exposed to EP were examined for cell cycle progression using flow cytometry, and DNA fragmentation was examined using the TUNEL assay. In addition, EP's mutagenic activity was assessed using the Salmonella typhimurium reverse mutation assay. The data showed that EP inhibits proliferation of MCF-7 cells, and it induces cell cycle arrest in G0/G1 through a DNA fragmentation-independent mechanism. Furthermore, EP's lack of overt cytotoxicity in the normal cell line HEK-293 and human fibroblasts in cell culture is supported by the absence of mutagenic activity of EP. CONCLUSION: EP emerges as a suitable molecule for further studies as a potential antineoplastic agent.

Los líquenes son producto de una asociación simbiótica entre un hongo y alga y/o cianobacteria; esta simbiosis produce una serie de metabolitos secundarios o sustancias liquénicas únicas, las mismas que son aisladas a partir de sus extractos y que han presentado una marcada actividad antibiótica y antifúngica. Considerando que en el Ecuador no se tienen antecedentes acerca de este tipo de estudios, el objetivo planteado fue el evaluar la actividad antifúngica del extracto liquénico de Usnea laevis frente a hongos fitopatógenos. En el presente estudio las sustancias liquénicas almacenadas en el talo de Usnea laevis fueron extraídas con metanol y etanol. La actividad antifúngica in vitro de estos extractos fue probado frente a géneros de hongos fitopatógenos previamente aislados (Aspergillus, Penicillium y Rhizopus stolonifer); añadiéndolo al medio de cultivo Agar Papa Dextrosa (PDA) a una concentración de 0,5%; determinándose el porcentaje de inhibición. Los datos fueron analizados estadísticamente y se realizó la clasificación del extracto en base a su porcentaje de inhibición según la OILB. El extracto metanólico y etanólico de Usnea laevis mostró una mayor efectividad frente a Rhizopus stolonifer, inhibiendo más del 50% de su crecimiento, a diferencia de Penicillium y Aspergillus cuyo porcentaje de inhibición fue mucho menor y mostraron diferencias estadísticamente significativas. El extracto liquénico fue moderadamente tóxico para Rhizopus stolonifer, ligeramente tóxico para Aspergillus sp 1 y Penicillium sp e inocuo para Aspergillus sp 2.


A lichen is an organism product of a symbiotic association between a fungus and algae and / or cyanobacteria; this symbiosis produces many secondary metabolites or unique lichen substances, which are isolated from their extracts and show a marked antibiotic and antifungal activity. Considering there is no background on this type of studies in Ecuador, the aim of this study was to evaluate the antifungal in vitro activity of Usnea laevis extract on phytopathogenic fungi. The lichenic substances stored in the thallus of Usnea laevis were extracted with methanol and ethanol. The in vitro antifungal activity of these extracts was tested against phytopathogenic fungal genera previously isolated (Aspergillus, Penicillium and Rhizopus stolonifer) . They were adding to Potato Dextrose Agar (PDA) culture medium at a concentration of 0.5%, and the inhibition percentage was determined. Data were statistically analyzed and the extract was classified based on its inhibition percentage according to the OILB. Methanolic and ethanolic extracts of Usnea laevis showed greater effectiveness against Rhizopus stolonifer, inhibiting more than 50% of its growth, unlike Penicillium and Aspergillus, whose inhibition percentage was lower and showed significant differences. The liquid extract was also moderately toxic to Rhizopus stolonifer, slightly toxic to Aspergillus sp 1 and Penicillium sp and harmless to Aspergillus sp 2.

ABSTRACT This study aimed to evaluate the teratogenic and hepatotoxic potential of the usnic acid encapsulated into PLGA-microspheres. In total, 12 female Wistar rats in pregnancy were randomly distributed in the control group (n= 6) that received 1.0 mL of physiological solution and treatment group (n= 6) that received 25 mg/kg of encapsulated usnic acid by oral administration. All females were euthanized at day 20 of pregnancy and their fetuses were removed and analyzed. During the pregnancy was observed a reduction in weight gain. There was no difference in serum transaminases levels analyzed as well as any difference in liver weight in both groups. The histomorphometric analysis of the liver from the treatment group revealed an increase in number of hepatocytes and a decrease in nuclear area of these cells. Moreover, no alteration was observed in cell area of hepatocytes or number of Kupffer cells. The fetuses had an increase in total number of hepatocytes and a reduction in the amount of megakaryocytes. These results show the hepatotoxic potential of usnic acid during pregnancy. However, its toxicity can be minimized by encapsulation in microspheres.

Resumen:La fragmentación de los ecosistemas es uno de los fenómenos antrópicos de mayor impacto global, y el efecto borde provoca que solo el interior de los fragmentos conserve sus características bióticas y abióticas originales. Los líquenes son organismos especialmente susceptibles a la variabilidad ambiental, lo que podría ser útil para la bio-indicación del efecto borde. En este trabajo se evaluó el efecto borde en dos fragmentos de Polylepis quadrijuga en el Páramo de la Rusia (Boyacá-Colombia), para determinar si existe un efecto borde sobre la distribución de los líquenes asociados a P. quadrijuga y su contenido de clorofilas. Se utilizaron tres transectos de 70 m a través del gradiente matriz-borde-interior en cada fragmento. Se escogieron nueve forofitos por transecto para medir variables ambientales: radiación fotosintéticamente activa, humedad relativa y temperatura del aire, y variables biológicas: riqueza y cobertura de cada especie. Además, fueron elegidas las especies que estaban presentes en las tres zonas del gradiente para cuantificar el contenido de clorofilas a y b, y determinar si hay cambios en la relación de clorofilas a/b que podrían sugerir plasticidad fisiológica como respuesta al efecto borde. Nuestros resultados mostraron que el fragmento 2 tenía una mayor exposición al borde debido a su alta relación perímetro/área, lo que causa una homogenización ambiental y pérdida de biodiversidad con relación con el fragmento 1. En general, se encontraron 55 especies que se distribuyen diferencialmente en relación con los fragmentos y el gradiente matriz-borde-interior. El interior del fragmento 1 fue la zona más conservada, que alberga una composición diferente en más de un 40 % a la composición de cualquier otra zona. Se clasificaron los líquenes de acuerdo con sus hábitos: gelatinosos, fruticosos, costrosos o foliosos, pero no se encontró ninguna relación entre la distribución por hábito y el efecto borde. Seis especies de amplia distribución mostraron cambios en el contenido de clorofila a lo largo del gradiente matriz-borde-interior, lo que es una evidencia de la plasticidad fisiológica al efecto borde. También fue posible diferenciar entre especies con preferencia por ambientes de alta temperatura y especies con preferencia a lugares más húmedos y suficientemente irradiados. Se concluye que algunas especies podrían tener un importante potencial como bio-indicadores de la fragmentación en ambientes de páramo.


Abstract:The ecosystems fragmentation is one of the anthropic phenomena with highest impact at global level and the edge effect causes that only the interior of fragments conserves their original biotic and abiotic characteristics. Lichens are organisms especially susceptible to environmental variability, what could be useful for bio-indication of edge effect. In this work, we evaluated the edge effect in two fragments of Polylepis quadrijuga in the Páramo de la Rusia (Boyacá-Colombia) to determine if there is an edge effect on distribution of lichens associated to P. quadrijuga and their chlorophyll content. We used three transects of 70 m across the matrix-edge-interior gradient in each fragment. We chose nine phorophytes per transect to measure the environmental variables: photosynthetically active radiation, relative humidity and air temperature, and the biological variables: richness and cover per species. Besides, we employed the species that were present in all the three zones of the gradient to quantify the content of chlorophylls a and b, and determine if there are changes in the ratio of chlorophylls a/b that could suggest physiological plasticity as a response to the edge effect. Our results showed that fragment 2 had a higher edge exposition because of its high relation perimeter/area, allowing to an environmental homogenization and lose of biodiversity in relation with fragment 1. Overall, we found 55 differentially distributed species in relation with the fragments and the matrix-edge-interior gradient. The interior of fragment 1 was the most conserved zone, harboring a composition different in more than 40 % to the composition of any other zone. We classified the lichens according with their habits: gelatinous, fruticose, crusty or foliose, but we did not find any relationship between the habit distribution and the edge effect. Six species of wide distribution showed changes in the chlorophyll content along the matrix- edge-interior gradient, what is an evidence of physiological plasticity to edge effect. It was also possible to distinguish between species with preference to warmer environment and species with preference to more humid and sufficiently irradiated places. Los líquenes son los hongos que establecen una relación simbiótica con un alga o cianobacteria. En esta simbiosis se producen por parte del hongo, una serie de metabolitos secundarios conocidos como sustancias liquénicas; las cuales presentan una marcada actividad antibiótica. En Cuba no se tienen antecedentes sobre estudios de metabolitos liquénicos por lo que se propone; evaluar el efecto fungicida de extractos liquénicos producidos por especies cubanas así como identificar sus metabolitos. Se emplearon líquenes de diferentes zonas del país (Parmotrema dilatatum, P. tinctorum, P. praesorediosum P. cristiferum, Ramalina americana, Cladonia ceratophylla y Cladonia portentosa spp. pacífica), a los cuales se les extrajo con acetona, las sustancias liquénicas almacenadas en el talo. Los extractos fueron probados contra los hongos fitopatógenos Rhizoctonia solani y Phythophtora nicotianae; por el método de envenenamiento del medio de cultivo agar papa dextrosa a concentraciones de: 0,01 por ciento; 0,03 por ciento y 0,07 por ciento. Se utilizó un control negativo de dimetilsufóxido al 0,07 por ciento y se determinaron los porcentajes de inhibición, cuyos resultados fueron analizados estadísticamente. Los metabolitos secundarios presentes en los extractos se identificaron por cromatografía de capa fina (TLC). Exceptuando el extracto liquénico de P. cristiferum, todos los demás mostraron más de un 50 por ciento de inhibición del crecimiento de ambos hongos a la concentración de 0,07 por ciento, mientras que a las restantes concentraciones los valores fueron variados con diferencias significativas con respecto al control. Se lograron identificar tres metabolitos liquénicos: metil 2‘-O- metilmicrofilinato, 4-O-Demetilmicrofilinico y el ácido ramaniloico.


Lichens are fungi that establish a symbiotic relationship with an alga or cyanobacterium. This symbiosis produced by the fungus, a series of secondary metabolites known as lichen substances; which show a strong antibiotic activity. In Cuba there is no background on the studies above lichen metabolites so it is proposed; evaluate the fungicidal activity of lichen extracts produced by Cuban species and to identify metabolites. Lichens from different areas of the country (Parmotrema dilatatum, P. tinctorum, P.praesorediosum, P. cristiferum, Ramalina americana, Cladonia ceratophylla and Cladonia portentosa spp. pacífica), to which it extracted with acetone, the lichen substances stored in tallus. The extracts were tested against the fungal pathogens Rhizoctonia solani and Phythophtora nicotianae; poisoning by the method of culture medium potato dextrose agar at concentrations of 0.01 percent; 0.03 percent and 0.07 percent. A negative control to 0.07 percent dimethylsulfoxide was used and the percentage of inhibition, the results were analyzed statistically determined. Secondary metabolites present in the extracts were identified by thin layer chromatography (TLC). Except P. cristiferum lichen extract, all others showed more than 50 percent growth inhibition of both fungi at concentration of 0.07 percent, while the remaining concentrations were varied values with significant differences from the control. Was made to identify three lichen metabolites metilmicrofilinato 2‘ methyl-O, 4-ODemetilmicrofilinico and ramaniloico acid.

A total of twenty four lichen species belonging to six families were collected from mountainous region of Nepal. The methanol extracts of each species were tested for antimicrobial and antioxidant activitiesin vitro. It was found that extracts of twenty one lichen species were active againstB. subtilis and seven species were active againstS. aureus. Similarly, in DPPH assay, three speciesPeltigera sp.,Cladonia sp., andCanoparmelia sp. showed comparable activity with commercial standard, BHA. In ABTS+ assay, extracts ofParmoterma sp.,Ramalina sp.,Peltigera sp. andCladonia sp. showed stronger activity than ascorbic acid. The observed data after comparison with previously published reports indicated that the high altitude lichens contain stronger antioxidant and antibacterial constituents. Similarly, the methanol extracts ofHeterodermia sp. andRamalina sp. showed comparable toxicity effect with commercial standard berberine chloride indicating a potent source of anticancer drugs.

The antischistosomal activity of the sulfated polysaccharide α-D-glucan (Glu.SO4) extracted from Ramalina celastri was evaluated after encapsulation into liposomes (Glu.SO4-LIPO) in Schistosoma mansoni-infected mice. The effect of treatment with Glu.SO4 and Glu.SO4-LIPO (10 mg/kg) on egg elimination, worm burden and hepatic granuloma formation was assessed using female albino Swiss mice, 35-40 days of age, weighing 25 ± 2 g, infected with 150 cercariae/animal (Biomphalaria glabrata, BH strain). Four groups (N = 10) were studied, two controls (empty liposomes and NaCl) and two treated groups (Glu.SO4-LIPO and Glu.SO4) using a single dose. Parasitological analysis revealed that Glu.SO4-LIPO was as efficient as Glu.SO4 in reducing egg elimination and worm burden. Treatment with free Glu.SO4 and Glu.SO4-LIPO induced a statistically significant reduction in the number of granulomas (62 and 63 percent, respectively). Lectin histochemistry showed that wheat germ agglutinin intensely stained the egg-granuloma system in all treated groups. On the other hand, peanut agglutinin stained cells in the control groups, but not in the treated groups. The present results suggest a correlation between the decreasing number of hepatic egg-granulomas and the glycosylation profile of the egg-granuloma system in animals treated with free Glu.SO4 or Glu.SO4-LIPO.

The aim of this study was to investigate metabolites of the lichen Laurera benguelensis. A high-performance liquid chromatographic (HPLC) method has been developed for the characterization of xanthones and anthraquinones in extracts of this lichen. Lichexanthone, secalonic acid D, norlichexanthon, parietin, emodin, teloschistin and citreorosein were detected in the lichen samples, which were collected from two places in Thailand. Components of the lichen were identifed by relative retention time and spectral data. This is the frst time that a detailed phytochemical analysis of the lichen L. benguelensis was reported and this paper has chemotaxonomic signifcance because very little has been published on the secondary metabolites present in Laurera species. Some of the metabolites were detected for the frst time in the family Trypetheliaceae. The results of preliminary testing of benzene extract and its chloroform and methanol fractions showed that all samples showed a weak radical scavenging activity. The chloroform extract showed the highest antioxidant activity.

RESUMO

Different nutrient elements were analyzed in the lichen Cryptothecia punctulata collected from the arecanut trees which were exposed to several sprays of a fungicide Bordeaux mixture. The study revealed the accumulation of fungicidal elements such as Cu, Ca and S in higher concentration of 575.4, 10,000 and 21,000 microg g(-1), respectively.

RESUMO

Lichens are lower group of plants. They accumulate heavy metals and have been used as bio-indicators in pollution monitoring. Nine lichen species were collected from urban, rural and hilly areas in the southern part of Tamilnadu. Accumulation of metals differed with species and also with respect to their locations. Among the nine species, Pyxine petricola collected from urban areas showed greater accumulation of the heavy metals i.e., copper, zinc, lead, nickel, chromium and cadmium.

RESUMO

Samples of foliose lichen, Dirinaria consimillis (Fr.) Awasthi were transplanted at six residential sites of Lucknow city for monitoring the concentration of Cr, Zn, Pb, Cu and Cd metals at two vertical positions. The concentration of chromium and copper were more at the higher vertical position than the lower one. The maximum zinc was at lower vertical position than the upper one. Lead and cadmium were not detected in the present study.

RESUMO

Para obter novos compostos com atividade antibacteriana, foram preparados os derivados 2,4-diidróxi-6-metilbenzoatos (orselinatos) e o ácido 2,4-diidróxi-6-metilbenzóico (ácido orselínico) através de reacões de alcoólise do ácido lecanórico. Todas as substâncias foram testadas contra bactérias Gram-positivas e Gram-negativas pelo método da microdiluicão. Staphylococcus aureus, Xanthomonas campestris var. vesicatoria and Ralstonia solanacearum foram mais sensíveis ao 2,4-diidróxi-6-metilbenzoato de n-propila, 2,4-diidróxi-6-metilbenzoato de n-pentila e 2,4-diidróxi-6-metilbenzoato de n-hexila com valores de CIM (Concentracão Inibitória Mínima) variando de 62,5 a 7,8 µg.mL1. Esses resultados mostraram que a homologacão da cadeia carbônica pode conduzir a compostos mais ativos.
